Question
Explain TWO of the following observations. Temperature-sensitive mutations, which cause proteins to denature at a lower-than nomal temperature, can promote amyloid formation by those proteins. a. b. An antibody binds the native state of a protein implicated in amyloid disease, the antibody appears to have therapeutic value for treating the disease. A molecular chaperone that assists protein folding can delay the onset of pion fomation in yeast. c. The ubiquitin-proteasome system that degrades misfolded proteins helps the cell minimize amyloid formation. d.Explanation / Answer
a. The amyloids are aggregates of proteins which are not folded in the required manner hence forming fibrils. If the protein denatures at lower temperature after mutation that means it has more tendency to lose it correct folding state as the denaturation temeperature has been lowered down, Hence more the denturation more are the chances of misfolding and aggregations.
c. Chaperones help in the folding of the proteins. prions are the infectious protein particles whic are formed by folding of proteins in multiple ways and bring the same channge in other proteins of same type. Now the presence of chaperons will bring specificity in the folding hence proteins will be folded correctly reducing the chances of prion formation.
