(A) The Sindbis virus infection was done in the drosophila flies with type w 1118 genotype and phenotype. The infection was monitored in the insect for 14 days and then the sample with the virus was collected and injected in fresh drosophila flies with w1118 infection. The comparison was made in the samples with luciferase counts in the negative and positive fractions of the infected flies along with the control studies.
(B) In the second experiment, the SINV virus infected sample was taken from drosophila flies infected with the this virus and mixed with the drosophila C virus and the pooled mixed sample with virus was injected in fresh w1118 strain of drosophila as compared to the control flies without virus infection. The sample was extracted from these flies after 4 days and luciferase activity was performed for the amount and intensity of infection. The amount and the count of infection with SINV and drosophila C virus increased as compared to the specimens with only SINV infection.
Hemocytes are phagocytic cells present in the hemolymph of the insects. These cells are like macrophages and ingest the double stranded RNA of the virus from the infected cells. The activity of the reverse transcriptase virus causes the formation of DNA strand from RNA.This DNA synthesizes the secondary viral siRNA(vsRNA) which interfere with the transcription of many genes inside the cell by blocking the mRNA formed inside the cell and preventing their translation by degrading the RNA. This complex includes the Dosha, miRNA, Argonaute complex which digest the mRNA of the genes formed inside the cell. These vsRNA are formed inside the infected cells of the insects and form an exosome complex. This sample is used to confer immunity in the animals by injecting the attenuated or dead samples containing this genetic material. The immune system of animals recognize these particles as foreign and synthesizes antibodies against these foreign viral protein particles with their inactive dead genetic material thus providing immunity to the animals.
