Question

The data shown were derived from a FRAP experiment on a transmembrane protein incorporated in vitro into liposomes made from a single, typical phospholipid species. The experiment was performed at 4 degree C. a. What is FRAP an acronym for? b. In one sentence or less, predict and explain how and why the results for this protein will change if the experiment was conducted at 37 degree Also, draw a solid curve on the figure above showing how the data would look. c. If you then kept the temperature at 37 degree C but incorporated cholesterol into your liposomes, what will happen? Predict and explain in one sentence or less, and draw a dashed curve on the figure showing how the data for this protein will change. d. Now, instead of liposomes you are monitoring the diffusion of this protein in the plasma membrane of living cells at 37 degree C. To your surprise the data look completely different (see Fig. B on right)! In 1-2 sentences provide a biological (not methodological) explanation.

Explanation / Answer

a) FRAP is Fluorescence Recovery After Photobleaching. It detemines the kinetics of diffusion through tissue or cells.

b) If the experiments are conducted at 37C, the result will change because, the protein activity will reduce at 37C and this temperature might also denature the protein.
