You're trying to count the persister cells in a culture. These are the cells that are not growing, but are not dead either. You have already counted the culutre with a Coulter Counter, and obtained a titer of 8.9 x 10^8 cells/mL in the culture. You should look at this as a two step process.

A. First, how would you figure how many cells in the culture were not growing?

B. Now, how would you figure out how many of the non-growing cells were still alive? Note that you can use any of the types of microscopes we discussed in Lecture 4 if you need to visualize the bacteria.

Explanation / Answer

non-growing but non-dead cells will deplet the oxygen present but at a slower rate because they need energy only to maintain their metabolism while dead cells will not deplet the oxygen content
