Results and Discussion The compounds anantxanthone and audracantone G ??0lated from Garona pauanens were found to confer pathogenic resistance in Nicotiana benthamiana to TMV. In Figure 1, Nicotiana benthamiana treated with ananixanthone, cudraxanthone G or water alone, were inoculated with infectious TMV sap and monitored for TMV induced- symptom developed for 6 weeks. Compare treatments and indicate which treatment(s) displayed strong resistance. Comment on how you would define resistance based on the figure presented. Both ananixanthone and cudraxanthone G are similar in structure, suggesting that one of these compound might be derived from the other by chemical or enzymatic means (Figure 2A). Compare cudraxanthone G verses ananixanthone and comment on extra ring structure and methoxy group (OMe). To examine ananixanthone and cudraxanthone G at the molecular level in relationship to TMV pathogenesis, Nicotiana benthamiana was treated with water, ananixanthone or cudraxanthone G alone followed by TMV inoculation. Figure 2B depicts TMV full-length genomic RNA and coat protein subgenomic RNA accumulation in leaves. Compare TMV full-ength genomic RNA and coat protein subgenomic RNA accumulation in the different treatments. Figure 2C depicts TMV coat protein accumulaton in leaves using an antibody specific for TMV coat protein. Compare TMV coat protein accumulation in the different treatments. In both Figure 2B and 2C slight differences in antiviral activities between ananixanthone and cudraxanthone G was observed. Which compound has stronger anti-TMV activity? What might be a reason why there is a difference in anti-TMV activity between the compounds? 05. Include the figures and figure textExplanation / Answer
1. Both treatments display strong resistance. In this case resistance can be defined as the maintenance of the integrity of the leaves because they do not dry up as compared to the water control in lane 3.
2. The structures of both molecules are indeed very similar. It is possible that by enzymatic reaction, the ring might be broken in one of the carbon links, the Oxigen in the OMe group was already present and the methoxy group is an CH3 which was also already present and it is simbolized in each corner of the ring. The difference between the two molecules is a simple breakage.
3 and 4. It appears that the treatment with ananixanthone erradicates more effectively the TMV organism, demonstrated here by the absence of its RNA and protein.
In conclusion, ananixanthone is a more effective treatment for TMV than cudraxanthone G.
