Question

Question 3 Consider the following data from Weinberg and Fersht (2005) J Mol Biol, 348:589 describing the binding of a protein ligand p53CT to a DNA receptor. The different symbols refer to different DNA sequences. Take the y-axis ('anisotropy") as an indicator of total observed binding. Focus on the trend that is common to all the curves, which represent a fit of the Hill equation to the data. At least how many sites does each DNA12 receptor have to bind p53? Are the sites interactive? If so, what type of cooperativity? What range in value does the Hill coefficient have? Explain. Explanation / Answer

Two receptor site must be bound with DNA, one is recognition site and other is pro-apoptotic site. Binding shows positive co-operative with lower affinity which fits to the Hill-plot which value ranges from -1 to +1. value of 1 shows high affinity positive cooperative binding and here p53CT ligand bind with high affinity to recognition receptor site which is involve in cell cycle arrest and -1 value shows positive co-operative binding with lower affinity indicated by -1 value.
