Question

2) A tumor specimen was isolated from a patient with late stage pancreatic cancer. You are given the task of identifying pancreatic cancer stem cells in order to determine their sensitivity to the chemotherapeutic agent paclitaxel. Previous studies suggest that pancreatic cancer stem cells express the cell surface markers CD44 and CD24 ) (24 pts) Describe four experiments (both in vitro and in vivo) that you should perform to identify and isolate the cancer stem cell population. For each experiment, describe how you will process the sample and illustrate the results that will indicate that you have isolated pancreatic cancer stem cells. Write your answers in the respective spaces below

Explanation / Answer

2.

Targeting cancer stem cell through CD133 and EpCAM/ESA could differentiate cancer cells from normal cells. It can help determine the sensitivity of the pancreatic skin cancer cells to paclitaxel as compared to the non- cancer stem cells.

The antigens to be studied for pancreatic cancer stem cells are CSCs, CD44, CD24 and ESA antigens through which overlap between the CD133+ and CD44+/CD24+/ESA+ cells in pancreatic cancer can be studied.
