Question

10,000 2- A molecule of DNA was digested with two restriction enzymes individually, (one enzyme in separate reaction, named enzyme 1 and enzyme 2, The samples were ran on an agarose gel. The order of loading samples are as follow (30 Points) Use graph and show the size of bands) how you estimate Iuncut DNA 2: cut DNA with restriction enzyme number 1 3: cut DNA with restriction enzyme number 2 4:DNA maker, containing S DNA fragments as follow 35 K bP, 15 Kbp, 15000 bp, 400 bp and 70 bp Answer the following question This DNA contained how many restriction site for enzyme 1 and enzyme 2? This molecule of DNA belong to prokaryotes or Eukaryotes, why What are the size of DNA fragments cut by enzyme 2

Explanation / Answer

1. The DNA contains one restriction site for enzyme 1 and three for enzyme 2.

2. The molecules belongs to prokaryote. If it is eukaryote it should be linear, it give two bands with enzyme I digestion, as it gives only one band with enzyme digestion 1 and also gives the same band with uncut DNA indicates it is circular DNA.

3. Plz do verify the fourth point because 15kbp = 15000 bp, then it will appear as same bands, but here they shown it as two diff bands. you can comment me with correct one...
