Question
For a phage titer of 3.3 x 10? PFUfm. and a bacterial títero?.6 x 10. CFU ml , calculate the multiplicity of infection (MOI) for 100 ? l of bacteria + 50 ?1 of phage. (1 pt) 9. 10 A researcher performed the transduction experiment with a set of bacteriophage and obtained the following data: PhageBacteriophage Bacteria titer Titer (pfu/ml)( MOI (25 ?1 phage + 100 ?1 bacteria) Number of | Transductants | | (25 ?1 of phage ise Transduction Percent Cells Transduced (cfu/ml) Frequency Alpha 5.7 x 10 Beta 3.9 x 10 Gamma 8.0 x 10113.0 x 10 135 45 178 5.0 x 10 4.0 x 10* From these data, determine the transduction frequency for each of the phage as well as the percent of cells that were transduced. 9) In this problem, concentration of phage is 3.3x 108 PFU/ml that can be written as
3.3x 108PFU/(1000ul) or 3.3x 105 PFU/ul
similarly we can write concentration of Bacteria = 1.6x1010CFU/ml = 1.6x107CFU/ul
Now, they have used 100ul of bacteria titer
so total number of bacteria = 1.6x107CFU/ul x 100ul= 1.6 x 105 CFU
and they have used 50ul of phage titer
so total number of phage= 50ul x 3.3x 105 PFU/ul = 16.5 x 104PFU
therefore, MOI= (total number of phage used)/(total number of bacteria used)
=(16.5 x 104 )/(1.6 x 105 ) = 1.03 phage/cell
