Explanation / Answer

1. Instead if water bath DNA is separated directly by using Lysate buffer. It lyse the cell and DNA is separated then.

2. Chloroform/phenol is used in 1:1 instead of protinase K. It inactivates and the proteins and other enzymes and also lyse the cell.

3. Salt in highconc. Precipitate the protein by salting out.

4. DNA is present in the supernatant and Cell debris is in the form of pallet. Supernatant is transferred to sterile centrifuge tube.
