Question

1. The Racker and Stoeckenius experiment confirmed the Mitchell Model of chemiosmosis. Which of the following is not true?
a. Bacteriorhodopsin contains all the components of the electron transport chain.
b. Bacteriorhodopsin pumps protons in the presence of light.
c. A proton gradient is sufficient for the production of ATP through the F1-F0 synthase.
d. The electron transport chain and ATP synthesis are separable.

2. True False  The 3' Ribose -OH is deprotonated and attacks the gamma phosphate of the incoming dNTP.

Explanation / Answer

1) Incorrect statement - D) Bacteriorhodopsin contains all the components of the electron transport chain.

The principle of this experiment is based on the fact that Bacteriorhodopsin does not contain the components of electron transport chain hence a proton gradient can not be generated by it's own mechanism. The proton gradient is thus created by Proton pump that are active in the presence of light and inactive in dark. This helped The Racker and Stoeckenius conclude that The electron transport chain and ATP synthesis are separable, as they could still produce ATP in Bacteriorhodopsin.

2) False)

The 3' Ribose OH is protonated as it has to acts a Nucleophile to attack the beta phospate of the next dNTP.
