There is an Ebola outbreak going on in Africa right now, and I volunteered my services to go test patient samples to determine who is infected. (If there was any doubt, this proves that I am a bit crazy.) We developeda branch chain technique for the identification of Ebola in patient samples. We ran a standard curve of Ong, 10ng, 20ng, 30ng, 40ng and got the following absorbance readings in order: 0,1.0, 2.0, 3.0, 4.0. We then ran 3 patient samples and got the following absorbance readings. Patient 1- 2.5, Patient 2- 0.5, Patient 3-3.5 a. (2 pts) What information would I need to give you for you to be able to tell me if we are testing for DNA, RNA, or protein? b. (6 pts) What is the concentration of virus in each patient? P1- P2_ , P3- c. (2 pts) Which patient do you think is at the latest stage of the infection?

Explanation / Answer

a. To know whether we are testing DNA, RNA or protein we need to know the absorbance value. Whether we ae measuring A260nm or A280nm. Generally the absorbance value of 260nm is used when we measure nucleic acids ( DNA or RNA) as the heterocyclic ring of nucleotides absorbs UV light at this particular wavelength. Whereas, to measure protein concentraiton absobance of 280nm is used. As amino acids contains aromati rings which absorbs UV light at 280nm.

    P3=35ng

c. pateint 2 is in the latest stage of the infection, as he carries less amount of EBOLA virus among the pateints.
