1)A) in this question , if you observe PCR gel, restriction enzyme digestion produce two frament in wild type, which similiar in unknown 2 means unknown 2 has nonsense mutation , in this mutation codon that code for amino acid change to stop codon but this change occur at protein level. Therefore, in northern blot there is no change but protein are truncated thats why western blot show protein of different molecular weight.

Unknown 3 show one fragment which means point mutation occur in gene X lead to missing restriction site means single fragment generated and thats why northern blot and western blot both show fragment of different size as compare to wild type.

Unknown 1also has deletion but it doesnt affect restriction site. So there is two fragment , one of large size and another is of same size of wild type.

B) unknown 2 and 3 has different phenotype than wild type due to mutation. Unknown 1produce protein fragment of same size as wild type and has a similiar phenotype like wild type
