Curare (tubocurarine) is previously used as an arrow poison as it can cause muscle paralysis. Curare is a type of non-depolarizing blocker of acetylcholine receptors in the skeletal muscle. Blockade of these receptors prevents the binding of acetylcholine, which is needed for the muscle depolarization that triggers muscle contraction.
At the neuromuscular junction, the nerve does not contain myelin sheath and gets expanded, which contains the stores of acetylcholine (Ach) in synaptic vesicles, which act as a neurotransmitter at the neuromuscular junction. Binding of Ach to the muscle receptor causes depolarization (influx of sodium ions) of muscle fibers and muscle contraction.
Curare acts by preventing the acetylcholine from binding to the receptor site, so it can be treated by using acetylcholinesterase (AChE) inhibitors such as "pyridostigmine."
