null mutation is a change in gene sequence so that it will either produce a non-functional protein or no protein production.
upstream of any ORF either insertion or deletion of a nucleotide will alter the sequence of the ORF. this will shift the ORF and it will end up with non-functional protein or no protein synthesis. ethidium bromide and proflavine are the intercalating agents which cause a frameshift mutation.
non null allele means functional gene. this is the mutation which does not alter the ORF of a gene. the gene product will be normal and functional.this mutation includes silent mutation, missense mutation (upstream of ORF).
