10. (apt) You are inoculating a KIA slant. Tell me what you sho 10 (1pt)You are inoculating a K?A slant. Tell me what you should do other than streaking the do a 11 ( pt). You have a sample that grew on BAP and Columbia agar, but not MacConkey. You of the blood around the colonies, it is catalase positive, and coagulase positive. What identify this bacteria as? Gram stain and find masses of purple cocci. Looking at your blood plate you see strong clea you 12. (1 pt). You have a bacterium that grew on BAP and Columbia agar, but not on MacConkey. Your On the BAP and Gram stain shows chains of purple cocci. The catalase test is negative. Columbia plates you note no distinct clearing of the blood and do Bile Esculin slants and Salt Broth. Your Bile Esculin Slant is mostly black and the Salt Broth is cloudy looking. What is it? 13. (1). You have a sample that grew on MacConkey agar and Blood agar, but not on Columbia agar. On MacConkey agar your colonies are bright pink. You do KIA slants and see a yellow butt and a yellow slant with a little red on the tip. You did some phenol red glucose and lactose broth indo tubes and both of those are yellow. There is no blackening of your KIA tube. For your le test you have red ring, the Methyl Red test gives you a red tube, and your VP does not look red. The Citrate tube is still green and Urea is a light salmon color. You did oxidase and found no blue color showing within a few seconds. What would you identify this bacterium as? 14. (1 pt). You have a sample that grew on MacConkey agar and Blood agar, but not on Columbia agar. On MacConkey your colonies are clear or yellowish. The oxidase test does not produce a blue color. You do a KIA tube and get a red slant and yellow butt, with some blackening in the butt. Your phenol red glucose turns yellow but phenol red lactose stays red. Your Indole test does not form a red ring, your Methyl Red test shows a red result and your VP test does not turn red. Your citrate is a blue color and your urea has turned bright pink. What is this organism? 15. (1 pt). You have growth on MacConkey agar and Blood agar, but not on Columbia agar. The colonies have a metallic sheen to them and an odd odor. You do the oxidase test off of the blood agar and get a dark blue color within seconds. You decide to do a KIA slant and get a red slant and red butt. You also do Fluorescent agar and get a blue color on top after incubation. What would you identify this bacteria as?

Explanation / Answer

Ans 10. Apart from streaking the slants, the slants should be stabbed with the same colony and should be incubated for 350C for 24 hours. After 24 hours the test would be interpretated for results.

Ans 11. Bacteria belonging to the staphylococcus species posses purple cocci and causes catalase and coagulase positive.

Ans 12. The characteristics would be observed in enterococci strains where blackenning of bile esculin slant and cloudy froth formation would occur.

Ans 13. Pink colour formation of macckonkey agar would occur due to present of certain lactose fermenters which are mostly enterobacteria. The rest other test such as the red coloured positive indole test would occur due to yhe presence of enterobacteria. Thus the present bacteria would be E.coli or other enterobacters.
