1. What is Gel Electrophoresis?

Ans: Gel electrophoresis is a technique which is used to separate the fragments of DNA, macromolecules such as RNA and Proteins.

Electric current is used to run these molecules on the gel made up of Agarose. DNA has negative charge on it. Hence, molecules move towards the positive electrode of the assembly. Large size molecules move slowly comparatively to smaller molecules

2. What is STR?

Ans: STR stands for Short Tandem Repeat Polymorphism.

The DNA strand has hundreds of repeated nucleotides. These nucleotides can be of 2 to 13 unit nucleotides in the single strand of DNA

STR Analysis can give the exact number of the repeating units. In this method, comparison of specific loci on the DNA from two or more samples.

3. Techniques used to determine the Paternity?

Ans: Paternity is done to prove that a person is the Father of a particular child

Techniques Used are:

? In this method, the analysis of various proteins and enzymes is carried out. Also the human leukocyte antigen (HLA) method is used.

The presence of antigen on red blood cells which are encoded by the ABO locus present on the human chromosome  

In this method the DNA sequences of both Father and child are compared and DNA profiling is carried out.

This method gives upto 99.99% accuracy.

4. What do restriction enzymes do?

Ans: Restriction Enzymes or Restriction endonucleases cleaves the DNA Strand at specific sites. It is a protein extracted from the Bacterial cells.

Restriction enzymes have property to cleave the Foreign DNA in the Bacterial Cells. They can be isolated from these cells.

Restriction enzyme recognizes a short, specific sequence of nucleotide bases (the four basic chemical subunits of the linear double-stranded DNA molecule—adenine, cytosine, thymine, and guanine).

These enzymes are very important tool used in the recombinant DNA technolgy in Genetic engineering
