Question
You set up the following gel in an attempt to construct a restriction map of the plasmid pUK1, using the restriction endonucleases Ban ll and Bg/ll. This series of questions is concerned with the principles of electrophoresis underlying this study. All digests were performed on circular DNA, which if uncut give no sixe information as they are a mixture of plasmids coiled by different amounts... Assuming that all wells received the same amount of DNA, Describe the band that contains the smallest amount of DNA.Explanation / Answer
(6)
Since the separation of DNA on the gel is dependent on the size of the DNA fragment, the smallest DNA in each lane are the bands towards the bottom of the gel (+ ve electrode end). The heaviest DNA will remain at the top of each lane.
