Due to nicotine, Ach is released and it binds to Ach binding protein (protein 1) - alpha subunit of cholinergic receptor - in response to which ion channels are open , permitting either one or more specific type of ions (K+ Na+ Ca++) to diffuse in to or out of the cell.
Ach when binds to protein 2 , muscarine Ach receptor - having 5 subunit, M1, M3, M5 receptor causes activation of phospholipase C - in response secondary messenger (IP3 and DAG) released - eventually leading to an intracellular increase of Calcium while M2,M4 inhibits adneylate cyclase, thereby decreaseing the production of secondary messenger cAMP . The activation of M2 receptor in heart is important for closing Calcium channel in order to reduce the force and rate of contraction.
