Question

3. Dr. Flora is studying the efects of high CO, levels on soybeans. She grew one 2 3 group of soybeans in regular air and another group of soybeans in air with high CO, levels. She labeled the cDNA from soybeans grown in high-CO, air with red dye and the cDNA from soybeans grown in normal air with green dye. Here are the results that she obtained from an experiment using a soybean gene microarray 4 5 6 Which spot or spots represent genes that were induced by elevated CO, Which spot or spots represent genes that do not show a difference in gene expression in high CO, levels vs. normal air. (Which sequences can Dr. Flora eliminate from further studies?) Explain your reasoning

Explanation / Answer

In microarray, several thousands DNA sequence is printed as tiny spot on microscopic slide. Now, samples from two different sources ( for eg, here soyabeans grwon on high CO2 and soyabean grown on low C02) were taken and mRNA is isolated. cDNAs are synthesized from the mRNA and each of them are tagged with fluroscence dyes ( like here red and green) and added to the microarry slide. Next, when the cDNA finds it complementary DNA sequence it will bind and from the appearence of red/ green spots we can identify which genes are expressed and which is not.

here in this experiment, cDNA isolated from syobean grown on high C02 is labelled with red dye and cDNA isolated from soyabean grown on nomal air is labelled with red. Thus genes which are highly expressed in high C02 will be expressed as red spots. Yellow spots are the genes which do not show any change as the mixture of red and green colour will turn yellow.
