Another student's unknown shows the following: a. Enterotube: Lactose is yellow, glucose is yellow with wax separated, H2S has no change. b. EMB: purple colony c. Predict TSI and include sugar fermentations: 6.

Explanation / Answer

Ans 1) Enterotube test is a multiple test system which was used to identify the enteric bacteria that is based on components like glucose, adonitol, lactose, arabinose, sorbitot and many other types of products. The possible reason why an organism or bacteria would not be identified through the test is its inability to ferment glucose.

Ans 2) A coliform bacteria is that which is rod shaped, Gram negative and non-spore forming bacteria. It is a motile or non-motile bacteria which has the ability to ferment lactose and as a result it leads to production of acid and gas. All the enteric bacteria are from the family Enterobacteriaceae and are able to ferment glucose to produce acidic end products. Coliforms are majorly ubiquitous in nature. The Enterococcus species are those which are gram positive cocci and pre-dominantly found in feces and is a human pathogenic bacteria.

Ans 3) Enterococci are gram positive, catalase negative cocci and is a part of normal intestinal flora present in humans and other animals. Yes, enterotube is the right method which helps in detection of Enterococi.

Ans 4) Yes it is possible for the bacteria to ferment lactose and not glucose depending on the enzyme it produces. Lactose is a complex disaccharide and only if the bacteria produces enzymes for its cleaving, it is able to ferment lactose and utilize it as energy. The bacteria that produces lactase is a able to ferment lactose.
