Question

PRE-LABORATORY ASSIGNMENT To be handed in to your professor before the start of lab. I. Name the four groups of macromolecules that are chemically digested within the alimentary canal. 2. Where is the major site of absorption throughout the alimentary canal? 3. What is the substrate for the enzyme salivary amylase? For pepsin? For pancreatic lipase 4. Name the protein, carbohydrate, and lipid substrates to be used in Exercise 1. Describe the appearance of a phenol red test of a sample where hydrolysis of the lipid substrate occurred to completion s. What test is used to determine if the carbohydrate substrate was hydrolyzed? What would be the appearance of a sample where there was an absence of hydrolysis? 6.

Explanation / Answer

1. Carbohydrates, proteins, lipids and nucleic acids.

2. Small intestine. Almost 90% absorption occurs in small intestine and rest 10% occurs in stomach and large intestine.

3. Amylase- carbohydrate; Pepsin- Protein; pancreatic lipase- lipids.

4. Any substrate can be used depends on availability. For example dextrose as carbohydrate, oils as lipids and milk as protein source.

5. Yellow, Hydrolysis of lipids produces fatty acids which make solution acidic. In acidic condition phenol red turns into yellow colour.

6. Inversion (hydrolysis) test is test is used to determine the hydrolysis of carbohydrates. Benedict's and Barfoed's ragent reacts with hydrolytic products of carbohydrates and gives positive results. In absence of hydrolytic reactions the colour of the solution is pale blue.
