Members of the epidermal growth factor receptor tyrosine kinase (RTK) family (EGFR/ERBBI, ERBB2/HER2, ERBB3/HER3 and ERBB4/HER4) are key targets for inhibition in cancer therapy. Critical for activation is the formation of a dimer in the cytoplasmic domain that induces autophosphorylation on specific tyrosine residues. A cytoplasmic protein, called MIG6, interacts with the cytoplasmic domains of RTKs and inhibits their kinase domain. Based on your knowledge of RTK signaling, what effect would you expect MIG6 overexpression in cancer cells to have on a) Ras activation, b) PI3-kinase activation c) MAP kinase translocation into the nucleus For each question briefly explain your answer.

Answer a. Based on the knowledge of RTK signaling, MIG 6 overexpression in cancer cell inhibit RAS activation. Since overactive Ras can lead to cancer and thus overexpression of MIG 6 inhibit Ras activation and cancer.

Answer b. Overexpression of MIG 6 also causes inhibition of PI-3 kinase activation. PI-3 activation pathway is important for cell proliferation and survival. MIG 6 also inhibit this pathway to stop cancer.

Answer c. MAP kinase translocation in to nucleus or MAP kinase pathway is important in protection of cell from stress and DNA damage. MIG 6 overexpression causes inhibition of MAP kinase translocation in to the nucleus thus causing DNA damage and inhibit cancer cell proliferation.
