Consider the following questions: why is the measurement of lung volume important to the diagnosis of both emphysema and lung fibrosis? Why is the measurement of collagen (hydroxyproline content) important in determining disease states in the lung? How would collagen content differ in fibrosis and emphysema? Finally, why do you think the authors needed to perform histologic examination of the lung tissue they had measured when the animals were alive?

1)The measurement of lung volume important to the diagnosis of emphysema and lung fibrosis because these volumes are calcuated based on stacked density histograms and showes good agreement with gold standard histopathology scores, total collagen content, these are longitudinal and quantitative follow up of dynamic changes in fibrotic and emphysematous pulmonary pathology. in fibrotic disease restiction of lung volumes is a hallmark features, these quantification of the aerated lung volume thus indirectly serves as an indicator of the extent of fibrosis. 2) The measurement of collagen is important in determing the disease state in the lungs because the validated ashcreft score was used to score pulmonary fibrosis , upon magnification of 100x each successive field was given a score ranging from 0(normal lung) to 8( total fibrous obliteration of the field). 3)The collagen content differ in fibrosis and emphysema , in lung fibrosis , the collagen is increased and in emphysema the collagen is degraded. 4)they developed and valiated a fully automated objective analysis protocol resulting resulting in quantitative volume and density data that corelated to the curent standard evluation techniques this approach opens new opportunities for the dynamic evaluation of pathological processes and putative therapeutic interventions within an individual animal.
