Explanation / Answer

A) In the presence of 2,4 dinitrophenol, electron transport from NADH to O2 proceeds in a normal fashion, but ATP is not formed by mitochondrial ATP synthase because the proton-motive force across the inner mitochondrial membrane is dissipated. This loss of respiratory control leads to increased oxygen consumption and oxidation of NADH. Hence, there is a large increase in the rate of O2 consumption when 2,4 dinitrophenol is added.

B) High concentrations of ADP are not inhibited by DNP. Instead they can be inhibited by AMP. DNP cannot be added instead of ADP because it acts as an uncoupler while ADP acts as a carrier. Without ADP no protons will be able to pass through the membrane and the experiment won't take place.
