In an effort to determine which ETC? component accepts electrons from reduced Vitamin C you decide to test the effect of adding (1) DNP, a H+ gradient inhibitor ( = an uncoupling agent) or (2) oligomycin, an ATPase inhibitor to the mitochondria along with reduced Vitamin. C, O2, ADP and Pi. A friend suggests that a better approach (3) might be take right-side-out IM vesicles and agitate them with glass beads then follow oxygen consumption in the presence of Vitamin C, O2,ADP and Pi. Note: Although not mentioned in class, whether or not IMM vesicles form inside - out ( = inverted) or right-side-out, depends on the buffer in which the mitochondria are disrupted (The reason for this is not understood). Thus both types of vesicles can be prepared. Which experiment (1). (2) or (3) would be the most likely to help you identify which of the two components of the ETC accepts electrons from Vitamin C. If you agitate the IM vesicles with glass beads it will cause the separation of the ETC complexes. thus, it can be found out which component of ETC accepts the electrons from vitamin C.
Because the other reagents affect the ATP synthesis process, i.e. DNP uncouples the electron gradient with H+proton gradient. and the role of oligomycin is to prevent H+flow back to matrix from the Fo component from the ATP synthase. thus, it can be said that the approach 3 is a better method.
