Question

9. An enzyme with a potentially useful application in synthetic biology has been discovered in a soil microbe. Unfortunately, this enzyme is not stable enough at the target temperature of 50 Degree C and on the timescale needed, as it tends to unfold with a time constant T unfold=400 sec. You have designed a mutant version of the enzyme with a few amino acid substitutions and found that it is significantly more stable than the wild type protein such that Double Delta G fold=10 kJ/mol. You also find that the mutation affects only the unfolding kinetics without altering the folding rate constant. What is the time constant of unfolding t unfold for your mutant protein at 50 Degree C?

Explanation / Answer

Time constant for unfolding remains the same i.e; 400 sec at the same temperature as the foldin kineticg and unfolding process is reversible. Only Unfolding kinetics changes.
