Gel type chromatography is also known as size exclusice chromatography (SEC) we use buffer solution in this to give iso-electric point of stabilization.This technique is used for purification of sampleof high weight contaminants.The mass of an unknown protein can be estimated by its elution position. As we lower the pH the net positive charge in the colunm decreases hence the column is negatively charged so so hace stability or to give iso-electric stability the protine with the net charge will bind with the coloumn but this process in unlike ion exhange chromatogaphy but the mass of an unknown protein can be estimated by its elution position where the protine is stuck in the bead.
