In lab, if you made an error and the solution was diluted to a smaller total volume than what was specified your absorbance measurement would [a]. decrease not change increase Why must you use the same cuvette (or test tube) for each sample on the Spec 20? Other cuvettes may be dirty and would alter the reading. A different cuvette would not fit into the instrument. The instrument is blanked for that cuvette, and other cuvettes are slightly different. There is no reason to use the same cuvette for each sample.

Explanation / Answer

1) Your answer is correct since Concentration = Moles / Volume. If you dilute less means volume is less hence concentration will increase for sure.

2) You answer is correct as each cuvette though looking same has some minor differences. When instrument is blanked for that cuvette it leads to the complete elimination of all the errors associated with that perticular test cuvette. Hence we must use the same cuvetter for all runs.
