Explanation / Answer

Mycobacterium can be isolated from mixed culture of bacteria in sputum by using solid culture or liquid culture medium. After that the culture is confirmed by the use of PCR technique.

Techniques used to isolate bacteria are:

1. Culture methods- These include culture on solid media, liquid media or using automated systems.

2. Non culture methods

Optimal atmospheric conditions, temperature, ph and vital growth requirements are needed for the bacteria to grow and isolate from the rest of the culture. Generally, 18-24 hours at 37oC are optimal for growth in most bacteria.

The staining techniques used to identify unknown sample are gram staining technique.

1. Gram positive bacteria are stained violet

2. Gram negative bacteria are stained pink

The staining protocols are as follows:

1. First the bacteria are stained with crystal violet dye

2. Then the bacteria is treated with mordant in order to fix the stain

3. The bacteria are then washed with alcohol which acts as a decolorizing agent

4. Finally safranin which is light red dye is used as counterstain

The gram positive bacteria retain the crystal violet stain while the gram negative are counterstained and give a pink appearance.

The main difference in the genera and staining is based on the walls of the bacteria. Gram positive have thick walls while gram negative bacteria have thinner walls.
