To test the function of a particular region of a protein the following approaches can be used.

a. The protein's domain (such as ATPase domain) can be mutated (replace a single or a stretch of amino aicds) and checked for its activity in vitro as in the case of enzyme function.

b.The protein domain (such as a Nuclear Localization Signal) could be mutated and episomally/stably/transiently/genome tagged expressed and checked for its activity or function in vivo.
