Question

1.

Medium #1

Medium #2

5 g yeast extract

10.5 g K2HPO4

20 g tryptone extract

4.5 g KH2PO4

0.5 g NaCl

1 g MgSO4

3.6 g glucose

10 g Polyurethane

1 l H20

1 l H20

Which medium would you consider to be Complex? Which medium is Defined? Which medium is Rich? Which medium is minimal? Explain your answers.

2. Why does growth of ETPUM in Medium #2 require oxygen? Think about this in terms of how ETPUM can generate a net gain in ATP by processing polyurethane. Remember that the degradation of polyurethane by polyurethanase does not expend ATP.

Explanation / Answer

Defined media is a kind of media that contains all the nutrients in known quantities and it doesnot have any nutrient whose components are unkown.

Minimal media contains minimum amount of nutrients and it supports growth of all kinds of microorganisms. Basically, it contains a carbon source such as glucose, succinate or polyurethane, salts containing essential elements to allow synthesis of Nucleic, protein and water.

Rich media supports growth of almost all types of microorganisms. It is composed of peptides or peptones (tryptone) which is used to synthesize amino acids, yeast extract to provide various organic compounds, vitamins, trace elements, minerals and sodium ions for maintaining the osmotic balance.

Complex media is a media that contains nutrients with unkown composition, for example blood agar media.

Here the medium 1 is a rich media as it contains all the nutrients which are present in the rich media and it is also the complex media since, the exact compostion of the yeast and peptone is not known.

Medium 2 is a defined media since the quantities of all the components are known. It is also the minimal media since contains polyurethane as a carbon source, salts and water.

Media 1 is Rich and Complex media.

Media 2 is Defined and Minimal media.
