Question

16 of 7 Column Chromatography HW 4. Student C needs to separate two compounds from one another and runs a column Student C then calls you over and explains that after running his column, he collected S fractions and he shows you the TLC of the fractionsfrom the colu an. Consider the following scenaios: 0.0 lic 1 2345 tu A. Student C loaded his sample onto the column diluted with too much solvent. Would this scenario lead to the above TLC result? Yes or No. Why? Student C used too nonpolar an eluent system when running the column. Would this scenario lead to the above TLC result? Yes or No. Why? B. Student C used too polar an eluent system when running the column. Would this scenario lead to the above TLC result? Yes or No. Why? C. D. Student C used too little adsorbent when running the column. Would this scenario lead to the above TLC result? Yes or No. Why? E. Student C added eluent prior to material adsorbed into silica when running the column Would this scenario lead to the above TLC result? Yes or No. Why? F. Student C used a column that had too large a diameter. Would this scenario lead to the above TLC result? Yes or No. Why? Copyright o 2016 Rochester Institute of Technology. Explanation / Answer

A) Such TLC results will not be obtained if sample is too diluted with solvent. It will just result in faster elution of sample on column hence difficulty in identification.

B) If too non polar solvent is used then it will not interact with the column as it is polar. We will not obtain such TLC pattern.

C) If too polar solvent is used then it will also not result in such a patttern. The solvent will show very high interaction with column.

D) Too little adsorbent is used It may result in the column as shown in the given figure. Because it will separate the compounds however the solute may run more of the column.

E) If eluent is added prior to material adsorbed then it will result in vague separation observed with a streak in spite of column. so such pattern as shown in figure will not be absrobed

F) If column with too large diameter is used then it will not result in such pattern as it will spread the solute and separation will not be regular.
