Question
Each of the following molecules violates Lipinski's rule of 5 for at least one reason. Indicate which reasons are violated for each molecule. If there is more than one answer, select all that apply o OO NH H OH 0 0 Ow OH OH 0 li = 365.51 log(P) 5.4 “ 695.80 = 462.41 log(P) 1.2 log(P) 2.7 Too many H-bond donors Too many H-bond acceptors Too big Too many H-bond donors Too man Too big Too many H-bond donors Too many H-bond acceptors y H-bond acceptors Too big Too hydrophobic Too hydrophobic Too hydrophobicExplanation / Answer
Lipinsk rule is a set of rules to determine wheather a molecule has potency of good drug. The rules are
1. There should be not more than 5 H - bond donors
2. There should be less than or equal to 10 H- bond acceptors
3. The molar mass should be less than 500 daltons
4. The octanol - water partition coefficient ( log P) should not be more than 5.
First molecule:
The first molecule has ( log p ) = 5.4 which is > 5 . It means it is too much hydrophobic .
For the reason this violets Lipinski rule
Second molecule:
It has 7 H bond donors ( all the H atoms attached to O or N are H - bond donors)
So; it violets the rule as it has too many H - bond donors
Third molecule:
It violets law as it has too many H bond acceptors ( all electronegative N or O atoms)
