Question

A student was asked to determine the protein concentration in a fraction saved during cytochrome c purification. She chose to use the Bradford method and used the standard stock BSA(0.05 mg/mL) to create a standard curve. She took 5 microL from her original sample and diluted it to a total of 1000 microL total. She used 10 microL from the diulted solution and added enough water and the dye to bring the total volume up to 3.0 mL and read the absorbance of the solution at 595 nanometers. The absorbance value was equal too 0.087. What is the protein concentration (mg/mL) in the original sample. The equation from the BSA standard curve is y=0.0264x+0.0129 where y is the absorbance and x is the concentration in micrograms/mL.

Explanation / Answer

y = 0.0264 x + 0.0129

y = absorbance

x = concentration

given the absorbance y = 0.087

y = 0.0264 x + 0.0129

0.087 = 0.0264 x + 0.0129

x = 2.807

concentration = 2.807 mg / mL
