Question
At locus VNTR63 there are 5 VNTR alleles (V63-1 to V63-5). Primers Left (PL) and Right (PR) are each 15-nts in length and are immediately adjacent to the VNTR repeats in the V63 region. Each VNTR63 repeat is 10-bp in length. A person with VNTR63-2 has two VNTR63 repeats in the region while someone who has VNTR63-4 has four VNTR63 repeats in the region . if there are 5 different VNTR alleles at the VNTR63 locus, then how many genotypes would be possible in the population (ie V63-1/V63-2 would be one genotype)? If you perform PCR analysis of this region using the PL and PR primers, how many bands will you see on a gel from the following genotypes?Explanation / Answer
There will be ten different genotype 5 alleles X 2 = 10
b. V63-2/V63-2 - Will have two bands since it has two VNTRs in the region
V63-4/V63-5 - Five bands- four for V63-4 and one for V63-5
V63-1/V63-2 - This will give three bands two for V63-2 and one for V63-1
C. The size of the DNA fragment will be 35 bp since V63 -4 has 10 bp, P left -10 bp and P right - 15 bp.= Total 35bp.
