Question
Use flybase (http://flybase.org/) as one of your references. Choose one of the following Drosophila genes to study: bom, tim or adh.
1. Describe the molecular role (if known) of the gene product
State a hypothesis to measure the phenotype(s) expected in the absence of the gene product.
2. Explain at least 2 different methods to mutate the specific gene you have chosen in Drosophila whole organism and/or specific tissues and study the function of the gene.
3. Describe the specific methods use on how you would determine that you have successfully knockdown, knocked out the gene’s expression at the RNA and protein level.
4. Describe at least 2 experimental approaches to test the phenotypes in the mutant tissues or whole organism. Include controls and numbers of samples/animals assayed.
5. For each experiment, justify why you might expect to observe certain results.
Explanation / Answer
Molecular role of bom gene product :- It is a family of 12 genes in Drosophila, known as Bomanins. The function is to provide immunity against microbial infection. These genes encode short peptides and the peptides are regulated by Toll pathway, related to innate immunity.
If the bom gene product is not present, then phenotype will be: Drosophila will not survive microbial infection, if these genes are not present.
