Question
As a researcher, you are asked to study the role of the App gene in mice.Use the Mouse Genome Informatics database (http://www.informatics.jax.org/) as one of your sources.
a. Describe the molecular role of the gene product
b. Compose a hypothesis regarding the testable phenotypes expected when the gene is knocked out.
c. Describe the method to use to knockout the gene.
d. Explain how you will verify that the genotype of the App knockout mouse is correct and how you will verify that reduced amounts and no App RNA and/or protein is being produced.
e. Design 2 experiments including controls to test for the presence of expected phenotypes.
f. Describe data collection methods and expected results.
Explanation / Answer
Molecular role of App gene:- App gene produces Amyloid beta A4 protein. It has a number of functions. It acts as a cell - surface receptor. It performs various functions on the surface of neurons and is related to neuron growth to a great extent.
When this gene is knocked out, neurite growth will be hindered.
By homologous recombination, the gene is knocked out. A 3.8 kb sequence encoding promoter and exon is removed. This is replaced by neomycin resistance genes.
