Introduction: Cytochrome P450 hydroxylates lipophilic compounds and the P450 monooxygenase system lacks substrate specificity.
Background: Allan Conney and others described that treatment of animals with different microsomal inducers like phenobarbital and 3-methylcolanthrene, results in different monooxygenase activity for metabolism of xenobiotics and steroid hormones. The species, diet, sex, age of animal are considered that are exposed to a chemical.
Purpose: To prove that there are multiple P450 enzymes with different substrate specificities in single microsomal extract.
Results: P450 catalytic profile alters in different membrane environment and there are multiple P450 enzymes with different substrate specificities.
Discussions: the multiple forms of P450s based on anomalies in P450 spectra were presented by Sato and Manning laboratories.
Conclusion: In two papers, which were published by Jud Coon in 1975 and 1976 confirms the presence of four isoforms of P450. He purified four isoforms of P450 from the rabbit liver.
