Question
You are given 10ml of a DNA stock solution that has 1.0mg target DNA/mL. The target DNA molecule is 2.9kb in length. Describe a set os serial dilutions that would yield a standard curve with the following concentrations: 2X10^7 copies/mL, 2X10^6 copies/mL, 2S10^5 copies/mL, 2X10^4 copies/mL. Make all of dilutions in water, and do NOT pipet less than 10mL. You may use as many tubes as you want for making dilutions.
I need the whole table filled out and none of the previous answers for this question were helping so if you can PLEASE fill out the table I would greatly appriciate it :)
Explanation / Answer
Average molecular weight of a nucleotide pair=660 daltons
2,900 base pairs weigh
= 2900*0.66
=1914 mg
In the given sample DNA has 6.02*1023 copies
Therefore,
10 mg contains
6.02*1023/1914*10
= 3.15*1021 copies
That is 3.15*1020 copies per ml DNA
M1V1=M2V2
M1=3.15*1020 copies per ml DNA
V1=10 mL
M2=2*107 copies/ml
V2=?
V2= M1V1/M2
= 3.15*1020*10/2*107
= 157,500,000,000,000
Take 10 ml of sample and dilute it in 157,500,000,000,000 ml of water to get 2*107 copies/ml sample.
