Gelatin hydrolysis is a test that is useful in separating the bacterial species of Clostridium, Bacillus, Pseudomonas, Serratia and Proteus.

MacConkey agar medium is used to isolate gram negative bacteria.

Mannitol salt agar medium is a selective medium that encourages the growth of gram positive bacteria.

Eosin methylene blue stain is used to stain gram negative bacteria.

SIM medium is used to detect the sulfide production, motility and indole formation.

Catalase is the enzyme that is released by the bacteria that can respire oxygen. The enzyme safe guards the organisms from the toxic by-products of oxygen metabolism. This test is used to differentiate catalase positive organisms from those of non-catalase. Aerobic and facultative anaerobes come under this category.

Facultative aerobes are organisms that can live in the presence of oxygen and can live even without it.

Facultative anaerobes are organisms that can live without the presence of oxygen and do not depend on oxygen for their survival. They can tolerate the presence of oxygen.

Aerotolerant anaerobes are the organisms that can tolerate the presence of oxygen and cannot use oxygen for their survival and growth.

Obligate aerobe is an organism that cannot live without oxygen.

Bacillus cereus is a rod shaped and is a gram positive bacterium. It is also catalase positive and grows only in the presence of oxygen. This bacterium is positive for gelatin hydrolysis test.

Escherichia coli is gram negative bacteria and are rod shaped. This is catalase positive and can live with or without oxygen. It is negative for gelatin hydrolysis. These bacteria did grow in Mac and SIM media and did not in MSA. They are stained by EMB.

Enterobacter aerogenes is gram negative bacteria and are rod shaped. This is catalase positive and can live with or without oxygen. It is positive for gelatin hydrolysis.

Enterococcus faecalis is gram positive bacteria and are chain shaped. This is catalase negative and cannot use oxygen for their growth though they tolerate presence of oxygen. It is negative for gelatin hydrolysis.

Lactobacillus plantarum is gram positive bacteria and are rod shaped. These are catalase negative and can live in the presence of oxygen. They are negative for gelatin hydrolysis.

Proteus vulgaris is gram negative bacteria and are rod shaped. They are positive for catalase enzyme activity and gelatin hydrolysis. They can live in the presence or absence of oxygen. They did not grow in MacConkey agar medium and can grow in SIM medium. They are not stained by EMB.

Pseudomonas aeruginosa is gram negative bacteria and are rod shaped. They are positive for gelatin hydrolysis and catalase enzyme. They did not grow in MSA medium. They cannot live in the absence of oxygen as oxygen is necessary for their survival.

Shigella flexneri are gram negative bacteria and are rod shaped. They are positive for catalase enzyme and negative for gelatin hydrolysis. They can live without the presence of oxygen. But, they can tolerate the presence of oxygen.

Staphylococcus aureus are gram positive bacteria and are spherical shaped. They are positive for catalase enzyme and gelatin hydrolysis. They did grow in MacConkey agar and MSA media. They can live in the absence of oxygen and can also live with it.
