Question
Practice solving the following solution problems: Every time you use Top Agar, you will need to supplement with CaCl2. How many ml of 100 mM Calcium Chloride are needed to add to 85 mL of Top Agar to have a final concentration of 1mNM CaCl2? 1. 2. How would you prepare 1L of 0.SM NaCl, using a 5M stock of NaCl and water? 3, How would you prepare a 40% glycerol solution, using undiluted glycerol stock and water? Here is a tough one to bring it all together. Phage Buffer is a reagent you will use almost every phage lab. Fill in the amount of each component in Phage Buffer to provide the proper final concentration to make 1 liter of phage buffer 4. Compound Stock concentration Desired Final Amount (mi) Tris buffer, pH 7.5 MgS04 NaCl CaCl2 ddH20 1 M 1M 5M 100 mM Concentration 10 mM 10 mM 68 mM 1 mMExplanation / Answer
I am solving the first question, Q1 as per Chegg guidelines, post multiple question to get the remaining answers
Q1)
Let the volume of CaCl2 added be x mL
The number of moles of CaCl2 will remain the same before and after the dilution
M1V1 = M2V2
M1 = 100 mM
V1 = x mL
V2 = (85+x) mL
M2 = 1 mM
x * 100 = (85+x) * 1
100x = 85 + x
99x = 85
x = 85/99 = 0.8585 mL
Hence we need to add 0.8585 mL of CaCl2 to 85 mL of agar solution
