Question

Describe the molecular changes expected to occur. Note whether the the mutation represents a gain-of-function allele or loss-of-function.

a. a mutation that makes it impossible to deoxyribonucleotides to bind to the polymerase active site of DNA polymerase I

b. a mutation in the coding sequence of DnaA in E. coli. The resulting DnaA protein can still bind DNA normally, but it can't bind at all to other DnaA proteins.

c. a mutation in topoisomerase II (DNA gyrase) that makes the enzyme unable to cut DNA

Explanation / Answer

a. a mutation that makes it impossible to deoxyribonucleotides to bind to the polymerase active site of DNA polymerase I
- Here elongation of the DNA strand doesnt take place, and if already the gene is replicated it will lead to gain of function, but in case if gene is not replicated it will lead to loss of allele function.

b. a mutation in the coding sequence of DnaA in E. coli. The resulting DnaA protein can still bind DNA normally, but it can't bind at all to other DnaA proteins.
- gain of function allele occurs because that one DnaA will bind at the oriC and unwinding of strand occurs, and replication proceeds.

c. a mutation in topoisomerase II (DNA gyrase) that makes the enzyme unable to cut DNA

- As the DNA cannot separate, the replication process does'nt takes place, which will lead to loss of function allele.
