Given the following scenarios name 2 methods that could be used to investigate these experimental situations and name the strengths and weaknesses of each. (Note there may be more than 2 techniques relevant to the scenario proposed.)
1) Chromosome Translocation: chromosome microarray + FISH? + CGH?
2) Determination of the levels of gene expression of Gene XYZ with or without hormone treatment in a cell line: FISH + (RT-PCR)
3) Presence of a K-ras mutation in a set of head and neck tumors: Sanger + Histology + NGS?
4) Gene Amplification: PCR + FISH? + qPCR
5) Identifying CD4 and CD8 positive T cells: Flow cytometry + immunohistochemistry
My answers are in bold, however, I'm not positive they're correct. If someone can please look at my answers and either confirm the accuracy or correct/add the right answers. Thanks so much in advance.

Answer
1) Chromosome microarray and FISH is the right technique but CGH is not preferred technique to detect chromosomal translocation.
2) RT-PCR is the technique which determines the level gene expression but FISH will not be useful to score the gene expression because It detects the DNA target in the chromosome but not the RNA or protein.
To find out the expression of gene one has to detect the level respective mRNA or protein encoded by that gene So right will RT-PCR and Western blot.
3) Sanger and NGS are right techniques to find out the mutation but not histology.
4) PCR and FISH are the right techniques to find out the gene amplification in the chromosomes
5) Flow cytometry and Immunocytochemistry rather than immunohistochemistry will be the right choice.
