Question

A student performed thin layer chromatography as described inthis experiment but made the following mistakes or changes in theprocedure. in each cases, explain how the change/mistake wouldeffect the Rf value: A. student failed to close the container contaninf thedeveloping solvent and in which the thin layer chromatography runwas carried out B. the solvent level in the developing jar was above theorginin line on the thin layer chromatography sheet C. student used as developing solvent, a mixture of twoimmiscible solvents D. student spotted too much of a concentrated sample of thecompound to be chromatographed E. the student dozed off and the solvent front ran off thethin layer chromatography plate before he stopped the run A student performed thin layer chromatography as described inthis experiment but made the following mistakes or changes in theprocedure. in each cases, explain how the change/mistake wouldeffect the Rf value: A. student failed to close the container contaninf thedeveloping solvent and in which the thin layer chromatography runwas carried out B. the solvent level in the developing jar was above theorginin line on the thin layer chromatography sheet C. student used as developing solvent, a mixture of twoimmiscible solvents D. student spotted too much of a concentrated sample of thecompound to be chromatographed E. the student dozed off and the solvent front ran off thethin layer chromatography plate before he stopped the run

Explanation / Answer

A) Elution won't be uniform. the solvent front gets dried often due to discontinuous elution. Rf obtained will be lesser than orginal.

B) Once dipping the spotted TLC plate inside the jar that contains more amount of solvent, then instead of elution the spotted sample started dissolving in the solvent.

C) Among two immisicble solvent, less densed solvent only will elute along TLC plate. Further uniform concentration won't be there if we use immissible solvents.

D) Steeking of sample will happen (no distinct Rf value)

E) Rf value will be more
