Question

Write the name of the most appropriate technique that you can to answer the question. Briefly explain (1-2 sentences) how this technique will answer the question. What DNA repair genes are expressed after exposing yeast to UV radiation? What DNA sequences does the engrailed transcription factor bind to in Drosophila? Does the amount of Hsp70 protein increase in E.coli following heat shock? You would like to examine the structure of the nuclear envelope, including the nuclear pores. What type of microscopy technique would you use and why?

Explanation / Answer

1a.The PHR1 gene of yeast encodes a photolyase enzyme which repairs specifically and exclusively pyrimidine dimers, the most frequent lesions induced in DNA by far-UV radiation. Therefore, PHR1 DNA repair gene is expressed after exposure of yeast to UV radiation.

1b. The engrailed trascription factor contains various lengths of upstreamed sequences. It is fused to a reporter gene coding for CAT( DNA) sequence in drosophila.

1c. Yes, the amount of Hsp70 protein increases in E.coli following heat shock.

During the heat shock response, E. coli expresses three major classes of heat shock proteins: Hsp70 (DnaK), Hsp60 (GroEL), and Hsp10 (GroES). The role of each of these proteins is to prevent or correct misfolded proteins, with Hsp60 and Hsp10 acting as molecular chaperones and Hsp70 acting to prevent aggregation of newly synthesized proteins, stabilize denatured proteins, and indirectly enhance the transcription rate of heat shock genes.

2. I would like to use high resolution scanning electron microscopic technique to examine the structure of nuclear envelopes with nuclear pores. Both the surfaces of nuclear envelope can be visualised in detail and more new information can be obtained.
