Many biochemists go bananas, and justifiably, when they see a Michaelis-Menten plot as shown above (ironically this figure is from the pearson publishing), in which the Vmax was under-estimated. To see why they are not happy, try to determine the V0 as a fraction of Vmax when the substrate concentration is equal to 10 Km. You will realize that when the substrate concentration equals 10 Km, Vo is only __________ % of Vmax (write down the whole number of the percentage, such as 80, 90, or whatever you get from your calculation below). So it is not appropriate to draw the Vmax line like the one shown above.

A + S = 10 KM, V0 =0.91 Vmax. I+S = 20 KM, V0 =0.91 Vmax.
