Question

3)TLC plates could be made of different particle size of silica. If one used a smaller particle size of silica in the TLC plate, how would that effect the results of a TLC experiment? Explain your answer.

4)the type of TLC performed in lab this week is often referred to as normal phase TLC. In a reverse phase TLC, the plate is coated with C18 hydrocarbon solid and the mobilephase used often consists of methanol and acetic acid or acetone. What would be the result if one used this system for separating the pigments of spinach? Explain youranswer.

Explanation / Answer

3) A smaller particle size of silica gel will cause a reduction on analysis time, the use of less amount of mobile phase, and higher precision on results. This is due, the smaller the particle size, the narrower the distribution, and more chance for the components adherent to the stationary phase in the mobile phase to adhere to the stationary phase. Also, as particles are smaller and they have a narrower arrangement, fluid travels easier through the stationary phase.

4) Hydrophobic pigments such as chlorophylls, carotene and xantophylls will attach to the now hydrophobic stationary phase, and hydrophilic pigments such as anthocyanin will move with the mobile phase.
