Question
You perform qPCR to determine expression levels of miR-102 in human multiple myeloma cancer cells (MM). After looking at the data (shown), you decide to look for targets of miR-102, and identify the following candidates based on sequence comparisons. Based on your data, and the information provided about each gene, write down if each gene is MOST LIKELY a target of miR-102, and explain each answer. a) Gene: Sithlord, Involved in cell cycle. Known to be overexpressed in MM cells b) Gene: Beiberkiller, Normally functions to inhibit cell division c) Gene: Plutoisaplanet, Is a pro-apoptotic gene in human cells d) Gene: Whobuysaminivan; Is an oncogene - increased expression in cancers e) Gene: Killmenow, Is a transcriptional repressor of WhobuysaminivanExplanation / Answer
A. gene sithlord- involved in cell cycle and known to be overexpressed in MM cells.
If sithlord were a target in miR-102, you would expect that it would decrease in cancer cells, since MM cells have higher level of miR compliment han normal cells and since sithlord show opposite phenotype. it not likely to be a target.
B. Gene Bieberkiller- normally function to inhibit cell division.
If BK is the target in the miR-102 we woukd expect low levels of BK in MM cells. Since or nomally inhibits cell division, low levels of BK should increase the cell div. which is likely observed in cancer cells. So BK is a target likely.
C. gene Plutoisaplanet- the proapoptotic gene in human cells
The proapoptotics genes act specifically on the abnormal/neoplastic cells causing their cell death.
D. Gene Whobuysaminivan- is an oncogen and show increased express ion n cancer cells.
This is also the target of miR-102 and should be lower in cancer cells. Since this gene is higher unlikely to act as a target.
E. gene Killmenow- is an transcriptional repressor of Whobuysaminivan.
High levels of miR-102 ans low levels of kmn. Since kmn is required for the expression of wbam, we would expect to see low levels of Wbam Gene in cancer cells. As Wbam is higher in the cancer cells, therefore, Kmn gene is unlikely to act as a target of miR-102
