Question

Below are 120 consecutive base pairs of DNA that include only the beginning of the sequence of gene X The underlined and italicised sequence (from position 15-40) represents the promoter for gene X The underlined and italicized sequence (from position 61-69) encodes the gene X ribosome binding (RBS) site. Transcription begins at and includes the T/A base pair at position 50 (bold). What are the first 3 nucleotides of the mRNA from gene X? Label the 5' and 3' ends of the sequence. What are the first 3 amino acids encoded by gene X? Label the N and C termini of the sequence. Design a forward primer that could be used as part of a primer pair to amplify this entire sequence. Label the 5' and 3' ends of the primer in your answer. Calculate the m of the primer. Show your working You have found cells with a mutation of gene X. The mutant cells have deletion of three base pairs (5' CTA 3) that includes the C/G base pair at position 80 (shown in bold) If the mRNA from this mutant gene can be translated, would you expect the protein to be longer, shorter, or the same as that produced from the normal gene X? If longer or shorter, indicate by how many amino acids.

Explanation / Answer

Termination codon does not carry the transcription but it terminates the process of translation because there is no tRNA having the anticodon to bind with termination codon. Hence the process of translation stops. Translation initiated with the help of start codon that is AUG. Small ribosomal complex bind with mRNA at 5’ end and recognizes the AUG codon from there they starts the translation.

In other hand the promoter does not helps in translation it only work for the transcription in which mRNA synthesized. At the promoter site the RNA polymerase will binds and starts the transcription.

Answer 1. 5’-CUA UCG GUC-3’ because these are the immediate sequences after the promoter sequences.

After transcription the translation will initiated.

Answer 2. After ribosome binding at specified site the ribosome will start translation from start codon which is present at 76th position 5’AUG.

N terminal (5’) – AUG ACU AGC- C terminal (3’)

Answer 3. Forward primer 5’-GGCTACTAATAAACTTGACAATAT-3’

                    Reverse primer 5’-CGGGGGCTGTACTAT-3’

Primer synthesised on basis of requirement. This set primer will amplify entire sequence. But that reverse primer included the repeated sequence this should be avoided.

Answer:- 4

Tm forward primer 49.86°

Reverse primer 47.51°C

Tm can be calculated using the formula:- 2 X (A+T) + 4 X (G+C)
